Eric Keller posts a 4 part series covering working with materials in ZBrush, starting with the basics and showing how to create your own standard materials, MatCap Materials, and Layering shaders, as well as LightCap and materials.

Eric provides some great tips in this series and he notes that if you are coming from V-Ray or mental Ray, you might be a little confused when working with standard materials in ZBrush. ZBrush standard materials are nowhere near physically plausible energy conserving materials.
